Most short river cruises in Porto depart from the Ribeira area or across the river in Vila Nova de Gaia. Always check your specific ticket for the exact pier.
Yes, especially during the summer and the September/October wine harvest. Wineries (quintas) often require advance reservations, and small-group tours sell out quickly.

The only thing that was disappointing is that we were unaware of the audio about the bridges that could be downloaded. More communication at the check in point would have been. Also the check in point was not easy to find.
A visit to Porto would not be complete without a ride down the Douro River. The views of the area and the six bridges are spectacular. This is a good way to put your feet up, relax and enjoy the city.
It was a beautiful boat, amazing hosts and views. Highly recommend! Went out to see the ocean and a few bridges. Didn't see all 7 bridges as the boat mast was too high to fit under a bridge. Sunshine, beautiful music, great hosts with stories. A lovely afternoon!
